put them as "tile.png" (or "tiles.png") in the mappack folder in "%appdata%\LOVE\mari0\mappack\custom_mappack_x"

I know this is old, but in case anyone comes here with a similar inquiry, Mari0 tiles require transparent pngs to create properties (AE has a workaround for this in the Animated Tiles) . I make all of my stuff using Aseprite, it's a very good tool for pixel art and sprites

paint.net lets you export transparent pngs also, as long as you're not saving it as a 24-bit image.
