Mario is the only playable character. Mario, Wario, Yoshi, and Luigi are now all playable.
In the game's opening and ending, Peach's hair is in its usual style. Peach is instead wearing a pony tail.
Mario was able to turn Invisible, become Metal, and Fly by collecting various caps. Mario can float, Luigi can turn invisible, Wario can turn Metal, and Yoshi can Breathe Fire by collecting a Power Flower, and Mario can fly by collecting a Cape Feather.
There are Blue, Red, Green, and Yellow ! boxes. There are Red ? boxes and Yellow ! boxes.
There are 6 missions in each portrait world (plus the 100 coin star.) There are 7 missions in each portrait world (plus the 100 coin star.)
There is one yellow Rabbit to catch twice. There are 7 multicolored Rabbits for each character to catch.
There are 120 Stars in the whole game. There are 150 Stars in the whole game.
Thwomps are blue and cube shaped. Thwomps are gray, spiked and rectangular prism shaped. Despite this, Mario can still stand on them.
Only Mario and Peach figures appear on the cake at the end. Wario, Luigi, and Yoshi figures appear along with those of Mario and Peach (with Wario and Luigi only appearing if the player has unlocked them).
After collecting all stars, the player may be shot out of a cannon to the top of Peach's castle, where they find Yoshi, who gives Mario 100 lives and a new animation for his triple jump. After collecting all stars, the player may be shot out of a cannon to the top of Peach's castle, but does not see Yoshi (unless they are playing as him). 3 1-Up Mushrooms can be found here, as well as one of Luigi's Rabbits.
Bubs and Bubbas appear. Bubs and Bubbas are replaced by Cheep-Cheeps and Big Berthas.
The Kuromames are black balls with no face. The Kuromames are replaced by motionless Fire Chomps.
Yoshi only appears at the end when the player is given 100 lives. Yoshi is the default player and the first to be played as.
The Super Mushroom did not appear. The Super Mushroom power-up was added, mainly found in bricks, and caused the player to grow to an enormous size (and gave Yoshi punching abilities). In the limited time one has it, one can defeat virtually every enemy, something one couldn't do in Super Mario 64.
Mario says "It's-a me! Mario!" on the Nintendo logo screen and "Hello!" on the title screen. Mario does not say anything on the Nintendo logo screen and says "It's-a me, Mario!" only on the title screen.
Mario is the only character featured on the title screen. Yoshi appears on the title screen if the player returns to it without turning the game off.
The analog stick is the only way to control Mario. As the DS does not have an analog stick, either the D-pad or the stylus can be used (depending on the settings). However, the slidepad on the Nintendo 3DS revives the classic analog stick feel.
Mario's face is pulled on in various places by the cursor. Mario's face or Yoshi's face is dragged by the stylus. While being dragged, they become wireframe. The user can also design a custom picture to drag around.
The C buttons control the camera. The L and R buttons control the camera.
Rabbits only provide hints and/or a star. Rabbits unlock minigames in the Rec Room at the Main Menu.
The door on the top right corner of the lobby only contains three Princess Peach murals inside it (one leading to The Princess' Secret Slide). The door on the top right corner of the lobby contains two Peach murals (one still leading to the slide), as well as five additional doors: the three doors with the letters above them allow the player to switch between characters; the remaining door contains a Power Star; and the door in the back center leads to the Rec Room.
Silver Stars are not present. Silver Stars were added; in these types of missions, Mario must collect five Silver Stars to make a Power Star appear.
Big Bob-omb has no particular interests. Big Bob-omb is obsessed with his mustache.
Mr. Blizzards have circular mouths and nothing on their head. Mr. Blizzards have zig-zag mouths and wear hats.
A glitch can cause Mario to Backwards Long Jump. Backwards Long Jumping is impossible.
The player always has the choice of playing any file. Only File A is available at the start.
Lakitu's Cloud resembles a normal, fluffy cloud. Lakitu's cloud isn't fluffy and has a happy face.
Lakitu looks unhappy. Lakitu looks happy.
When a file begins, Mario immediately says "Ha Ha!" after saying "Yahoo!" When a file begins, Mario pauses before saying "Ha Ha!"
All crates are brown. Some crates are brown, while others are black. Black crates can only be broken by Wario.
